Questions producers ask

Does the kick end up in the bass stem?

No. The kick stays in the drum stem. The bass stem carries the bassline itself, including the sub content.

Can I detect the notes from the stem?

Much more easily than from the full track. Run the isolated stem through your DAW's audio-to-MIDI and the results are dramatically cleaner.
